Can I do this with DvdProfiler?
1.  Open dvd storage drawer #1.
2.  Remove dvds and scan sequentially with handheld barcode scanner.
      The software recognizes each new barcode as a new entry without any user intervention.
      The storage location code field defaults to the location entered for the first entry.
3.  Replace dvds and close drawer #1, proceed to drawer #2, changing location code.

1. No, DVD Profiler can't open your drawer, but there is a plug-in available for that called The_User ... 

2. Well exactly in that way it is not possible, but: You can scan all the barcodes into one simple textfile (or rather one textfile per drawer) and afterwards DVD -> Add to Collection -> Add by UPC -> Add Multiple and open the texfile.

The Location is a bit trickier as unfortunately there is no function in DVD Profiler (or I have overlooked it) to copy this information from one profile to another en mass. If you just want the Location "Drawer #1" I suggest to use a Tag to preserve this info and later on when you are comfortable with the program install and use the Plugin BulkEdit to enter this data in it's proper fields.
For the Tag: after importing flag all the profiles by hitting space on each or Ctrl&a for all visible, Collection -> Flagged -> Set Tags and click "Drawer #x"

3. repeat the steps in 2. and for the tagging: Ctrl&f to open the filter-query, double-click "Drawer #1" to hide all items already belonging to it and for the rest repeat: Ctrl&a for all visible, [i]Collection -> Flagged -> Set Tags and click "Drawer #x"[/i]

PS As an unregistered user you have almost all the features available to you as long as you have under 50 profiles, so give it a nice testdrive around the block.
