Roy viser sig at være mesterspion, så hun kommer på sit livs eventyr. Roy har brug for Junes hjælp til at beskytte et objekt af helt afgørende betydning for den globale sikkerhed. På en rejse, der går fra Wichita lufthavn til Boston, videre til en fjern paradis-ø og ender i Sydspanien, gør June og Roy alverdens krumspring for at undvige de lejemordere og agenter, der er lige i hælene på dem. Midt i de konstant skiftende alliancer og uventede forræderier i 'Knight and Day' forsøger June at gennemskue, hvorvidt Roy er hendes drømmemand eller i virkeligheden en skør, paranoid landsforræder.
